poweroff the vm
in Virtualbox > Settings > Display > Monitor count > 2
start VM then View -> Virtual Screen 2 -> Enable
does this help?

before this step, did you configure the guest to extend the desktop to a second monitor?
(I assume you have guest additions installed)

Is your virtualbox Graphics Controller VBoxVGA ?
and extend the video memory to 128M
It's working here on a Manjaro host:
The second monitor was avaliable as soon as it logged on to the desktop. In this VM I never attempted to install from Ubuntu's Additional Drivers and used it straight off from the Guest Additions CD.
I have found Guest Additions 6.1.4 to be buggy on 20.04 at the moment - shared clipboard doesn't seem to work!
Just to note, VirtualBox 6.1.6 has been released and addresses this area:
